Novel ini juga menyajikan kebiasaan konsumsi narkoba SHerlock Holmes dan memanusiakannya dengan cara yang belum pernah dilakukan dalam novel sebelumnya. Saat itu, Dr. Watson memperkenalkan calon istrinya kepada Holmes, yang bernama Mary Morstan. Mary pada saat itu menceritakan kepada Watson dan Holmes tentang ayahnya. Ayahnya pernah mengirimnya telegram setelah kembali dari India. Ayahnya meminta Mary untuk menemuinya di Hotel Langham, London. Namun sayangnya, ketika tiba di hotel Mary diberitahu bahwa ayahnya pergi dan belum kembali ke hotel sejak semalam. Mary pun kebingungan dan menghubungi salah satu teman ayahnya yang bernama Mayor John Sholto. Holmes langsung mengerjakan kasus tersebut dan menemukan fakta bahwa Mayor Sholto sudah meninggal enam tahun lalu. Di saat yang bersamaan,Mary menerima mutiara-mutiara dari pengirim anonimus. Mary pun memberikan petunjuk lain kepada Holmes yaitu sebuah peta yang ditemukan di meja ayahnya. DI peta tersebut bertuliskan nama-nama asing seperti Jonathan Small, Abdullah Khan, Mahomet Singh dan Dost Akbar. Holmes, Watson dan Mary akhirnya memutuskan untuk bertemu anak Mayor Sholto, Thaddeus Sholto. Setelah ditelusuri, ternyata dia adalah pengirim mutiara anonim itu. Namun, sang anak mengakui bahwa ayahnya melihat ayah Mary pada malam dia meninggal. Pada malam menjelang kematian, Mayor Sholto mengakui bahwa dia telah membunuh Morstan dan memberitahukan letak harta karun yang disembunyikan. Novel ini menceritakan sebuah kisah tentang percobaan pembunuhan. Kisah ini terinspirasi dari legenda anjing buas yang sangat kejam dan menakutkan. Sherlock Holmes dan Dr. Watson menyelidiki kasus ini bersamaan. Pada buku ketiganya ini menjadikan cerita Sherlock Holmes yang paling terkenal yang pernah ditulis oleh Conan Doyle. Dia pun juga mengkhawatirkan keturunan Baskerville berikutnya yaitu Sir Henry Baskerville. Meskipun Sherlock Holmes tidak terlalu percaya dengan kutukan omong kosong itu, ia tetap menemui Sir Henry Baskerville. Dia adalah seorang pemuda tampan nan periang, dan sangat skeptis tentang legenda Baskerville itu. Dia pun mengaku bahwa dia juga pernah menerima catatan dari anonim yang memperingatkan dia untuk menjauh dari rawa-rawa. Di antara perkebunan yang luas itu juga terdapat pasir hisap yang bisa merebut nyawa hewan maupun manusia. Bahkan pernah ada berita seorang narapidana yang melarikan diri dan bersembunyi di area perkebunan tersebut. Sir Henry dan Watson tidak bisa tidur saat malam hari. Mereka hanya bisa bersantai pada siang hari dan menjelajahi lingkungan sekitar serta bertemu dengan penduduk di sana. Watson terus berusaha mencari petunjuk tentang siapa orang yang telah mengancam Sir Henry Baskerville. Ia juga tidak lupa selalu mengirimkan laporan kepada Holmes. Cerita di dalam novel ini pertama kali diterbitkan di sebuah majalah bernama Strand Magazine mulai dari September 1914 sampai Mei 1915. Tidak seperti novel-novel sebelumnya, pada novel keempatnya ini Conan Doyle menuliskan dua bagian dengan hati-hati yang dipenuhi dengan ketakutan dan teror. Novel ini juga mengangkat tema utama yaitu masalah etika dan aktivitas teroris oleh serikat pekerja Amerika. Novel yang diadaptasi ke dalam Film dan serial TV ini menceritakan kisah Sherlock Holmes yang menerima sebuah pesan berisi sandi dari Fred Porlock, seorang agen dari Profesor Moriarty. Namun, Porlock berkhianat kepada Profesor Moriarty dan dia takut bahwa pengkhianatan itu diketahui oleh Moriarty. Dia akhirnya menceritakan kepada Holmes perihal hal ini. Dari pesan sandi itu Holmes menyimpulkan bahwa sandi buku yang digunakan untuk enkripsi adalah buku biasa yang besar,berisikan 534 halaman. Holmes pun mencoba buku edisi terbaru Whitaker’s Almanack, namun gagal. Dengan memecahkan sandi tersebut, holmes berhasil menguraikan pesan yang berisikan peringatan rencana jahat terhadap seorang pria yang tinggal di Birlstone House bernama Douglas. Tidak berselang lama setelah Holmes berhasil memecahkan sandi tersebut, Inspektur MacDonald mendatangi kediaman Homes. Ia memberitahukan bahwa Douglas telah dibunuh. Holmes memberitahu bahwa kejahtan ini ada keterlibatannya dengan Moriart. Namun Inspektur MacDonald tidak mempercayai bahwa orang berpendidikan dan terhormat seperti Moriarty melakukan tindakan keji seperti itu. Akhirnya, Inspektur bersama Holmes dan Watson pergi ke Birlstone House, untuk menyelidiki kejahatan tersebut. Douglas ditemukan sudah terbaring di ruang tengah dengan sebuah senapan gergaji yang terletak di atas dadanya. Dia telah ditembak oleh seseorang dari jarak dekat, kepalanya hancur berkeping-keping. Selain itu juga ditemukan kartu di samping tubuh Douglas dengan inisial dan nomor 341. Holmes dan Dr. Watson sampai di lokasi pembunuhan dan mendiskusikan kasus tersebut dan setuju bahwa itu bukanlah kasus bunuh diri. Douglas diketahui merupakan seorang buron dan pindah ke pedesaan Inggris. Ia menikah di Inggris, namun istri pertamanya telah meninggal karena penyakit tipes. Selain itu diketahui juga oleh temannya Douglas, Barker, bahwa Douglas pernah menyebutkan sesuatu tentang The Valley of Fear’. Holmes dan Watson pun memutuskan untuk mencari penginapan guna melanjutkan penyelidikan besok. Sesampai di penginapan, Holmes memberitahu Watson bahwa Barker dan istri Douglas sudah berbohong. Selain itu, Holmes juga mengetahui bahwa pengurus rumah itu mendengar suara seperti pintu yang dibanting. Holmes pun berasumsi bahwa itu adalah suara tembakan dari senapan. Awalnya mereka mengira orang Amerika yang menjadi pembunuh Douglas, namun mereka tidak menemukan bukti-bukti yang kuat. Akhirnya, Holmes pun menulis surat kepada Barker untuk mencari sesuatu di sekitar parit. Holmes, Watson dan Inspektur Mcdonald melihat Barker di parit. Akhirnya Barker pun disergap oleh mereka. Namun siapa sangka, Douglas yang dikiranya terbunuh itu muncul dalam keadaan baik-baik saja. Dia akhirnya menjelaskan kepada Watson dan Holmes tentang The Valley of Fear’ yang menjelaskan tentang mengapa dia takut akan kehidupannya.
